Multi-Faceted Roles of ERCC1-XPF Nuclease in Processing Non-B DNA Structures

چکیده
Genetic instability can result from increases in DNA damage and/or alterations repair proteins and contribute to disease development. Both exogenous endogenous sources of structure (e.g., non-B DNA) impact genome stability. Multiple mechanisms exist counteract damage. One key protein complex is ERCC1-XPF, a structure-specific endonuclease that participates variety processes. ERCC1-XPF involved nucleotide excision (NER), interstrand crosslinks (ICLs), double-strand break (DSB) via homologous recombination. In addition, contributes the processing various alternative (i.e., non-B) structures. This review will focus on structures by ERCC1-XPF.

Reconstitution of human excision nuclease with recombinant XPF-ERCC1 complex.
The human XPF-ERCC1 protein complex is one of several factors known to be required for general nucleotide excision repair. Genetic data indicate that both proteins of this complex are necessary for the repair of interstrand cross-links, perhaps via recombination. To determine whether XPF-ERCC1 completes a set of six proteins that are sufficient to carry out excision repair, the human XPF and ER...
متن کاملMultiple roles of the ERCC1-XPF endonuclease in DNA repair and resistance to anticancer drugs.
In this review, we focus on the discrepant roles of the DNA repair complex ERCC1/XPF in the prevention of cancer and in the resistance of cancer to chemotherapy. ERCC1/XPF is essential for nucleotide excision repair (NER) incising DNA 5' to the lesion. NER deficiency results in the skin cancer-prone inherited disease xeroderma pigmentosum (XP). The ERCC1/XPF complex is also involved in recombin...
متن کاملUSP45 deubiquitylase controls ERCC1–XPF endonuclease-mediated DNA damage responses
Reversible protein ubiquitylation plays important roles in various processes including DNA repair. Here, we identify the deubiquitylase USP45 as a critical DNA repair regulator. USP45 associates with ERCC1, a subunit of the DNA repair endonuclease XPF-ERCC1, via a short acidic motif outside of the USP45 catalytic domain. Wild-type USP45, but not a USP45 mutant defective in ERCC1 binding, effici...
متن کاملERCC1-XPF endonuclease facilitates DNA double-strand break repair.
ERCC1-XPF endonuclease is required for nucleotide excision repair (NER) of helix-distorting DNA lesions. However, mutations in ERCC1 or XPF in humans or mice cause a more severe phenotype than absence of NER, prompting a search for novel repair activities of the nuclease. In Saccharomyces cerevisiae, orthologs of ERCC1-XPF (Rad10-Rad1) participate in the repair of double-strand breaks (DSBs). R...
متن کاملMouse SLX4 Is a Tumor Suppressor that Stimulates the Activity of the Nuclease XPF-ERCC1 in DNA Crosslink Repair
SLX4 binds to three nucleases (XPF-ERCC1, MUS81-EME1, and SLX1), and its deficiency leads to genomic instability, sensitivity to DNA crosslinking agents, and Fanconi anemia. However, it is not understood how SLX4 and its associated nucleases act in DNA crosslink repair. Here, we uncover consequences of mouse Slx4 deficiency and reveal its function in DNA crosslink repair. Slx4-deficient mice de...
